HEXB promoter induces reporter transcription mainly in human microglia and not in astrocytes. Native mouse, human, or monkey HEXB promoter-driven dual-LG reporters were transfected into human primary microglia (A,B) or astrocytes (C,D) plated in 96-well plates. After transfection, the presence of GFP+ cells was determined by fluorescence microscopy (A,C) where the transfection efficiency was determined by flow cytometry (CMV-LG or CMV-GFP). gdLuc levels (B,D) in culture media were used to determine the activity of the indicated HEXB promoters.
