(A) Antigen-specific T-cell-derived T-iPSC differentiates into TCRαβ T cells, which partially lose antigen-specificity resulting from mispaired endogenous TCR expression. (B) Knockout of RAG2 at T-iPSC stage eliminates the additional TCR rearrangement and preserves the antigen specificity of induced T cells from T-iPSC. (C) Myeloid-cell-derived non-T-iPSC are transduced with antigen-specific TCR and differentiate into antigen-specific induced T cells with rare activation of endogenous TCR rearrangements.
