Design of the Kv8.2 KO strain. (A) Kv8.2 (red) requires assembly with Kv2.1 (gray) to form a functional channel. Kv8.2 has the typical topology of a voltage-gated K+ channel subunit; a T1 tetramerization domain, voltage sensor and pore-forming domain. Guide RNAs were designed to excise 101 nt from Kcnv2 to generate a frameshifting truncation in the N-terminus of Kv8.2 prior to any functional domains.
