ICN 17261 and ribavirin both suppress hepatic injury in a murine hepatitis model measured using serum ALT as a surrogate marker. Three groups of six mice were injected i.p. with 1-mg/kg ICN 17261 (Con A + 17261) 1-mg/kg ribavirin (Con A + RIB), or PBS (Con A + PBS) 1 h prior to intravenous injection of 0.3 mg of ConA. Twenty-four hours later, all mice, including an untreated group, were exsanguinated, and serum ALT levels were determined by using a colorimetric assay. Data were obtained as absorbance values (A505) ± SD and are representative of three separate experiments. Serum ALT concentrations were calculated from a calibration standard curve (Sigma) and were converted to international units per milliliter.
