DCHP exposure does not affect the migration and adhesion properties of macrophages of PXRF/FLDLR−/− and PXRΔMyeLDLR−/− mice. Four-week-old male PXRF/FLDLR−/− and PXRΔMyeLDLR−/− littermates were treated with 10 mg/kg/day of DCHP or vehicle control daily by oral gavage for 12 weeks. (A) Peritoneal macrophages isolated from the PXRF/FLDLR−/− and PXRΔMyeLDLR−/− mice were labeled with calcein acetoxymethyl and cultured with primary porcine endothelial cell monolayer for 30 min. Adhered cells were then counted under a fluorescence microscope (scale bars = 100 µm). (B) Peritoneal macrophages isolated from the PXRF/FLDLR−/− and PXRΔMyeLDLR−/− mice were seeded on the Matrigel-coated transwell filters. Cells that infiltrated and migrated to the underside of the transwell were stained with hematoxylin and counted under the microscope (scale bars = 100 µm). Quantitative analysis of the migrated and adhered cells was displayed below the representative images. Data are represented as mean ± SEM (n = 3).
