Kaempferol reverses the 5-Fu resistance in HCT8-R cells. (A) HCT8-R and FHC cells were treated with increasing concentrations of kaempferol for 48 h, and the cell viability was determined by CCK-8 assay. The blank control group (0 μM) represented 0.05% (v/v) DMSO (dimethyl sulfoxide). Data represent means ± SD from three independent experiments. (B) HCT8-R cells were seeded into a 6-well plate at a density of 5 × 103/well and treated with various doses of kaempferol for one week; then the cell clone formation ability was explored. (a) Representative graphs are shown. (b) The quantitative analysis of colony formation. (C) Analysis of apoptosis in HCT8-R cells treated with kaempferol for 48 h using flow cytometry. (a) Representative graphs are shown. (b) The apoptotic rate is shown in bar format. (D) HCT8-R cells were pretreated with different concentrations of kaempferol for 24 h, then treated with 5-Fu for another 24 h, and the cell survival rate was determined by CCK-8 assay. * p < 0.05, ** p < 0.01.
