Phagocytic and chemotactic ability of PMs from LTM and p190RhoGEF TG mice. PMs from LTM and TG mice were cultured in media with or without LPS (100 ng/ml) overnight. These PMs were used for the assay. (A) PMs (2.5 x 105) were incubated with FITC-conjugated zymosan (20 μg/ml) at 37°C for 30 minutes. The PM population made fluorescent by zymosan internalization was detected using flow cytometry. The data are the mean ± SEM of five independent experiments. Statistical significance was analyzed by one-way ANOVA with Tukey’s multiple comparison tests. Significant comparisons are noted as **p < 0.01. (B) PMs (2.5 x 105) were added to the upper wells of a Transwell plate that contained the chemoattractant fMLP (1 mM) in the lower chambers. The cells that migrated to the lower chamber were counted after the plate was incubated at 37°C for 2 hours. The data are presented as the percentage of the total cells that migrated. The data shown are the mean ± SEM of three independent experiments with duplicate sample settings. Statistical significance was analyzed by one-way ANOVA with Tukey’s multiple comparison tests. Significant comparisons are noted as *p < 0.05, **p < 0.01.
