Pharmacokinetics of Fc variant antibodies. Homozygous Tg32 mice (n = 4 for wild-type (WT) and acid-Fc; n = 3 for YTE, female and male) expressing the human FcRn under the human promoter were administered 2 mg/kg of each antibody intra-peritoneally after which tail vein samples were collected every ∼3 days. Serum concentrations of the administered human antibody were determined by antigen-specific ELISA and plotted against time to determine the beta elimination rate constant β. Serum elimination half-lives were determined from the measured rate constants as t_1/2β = ln2/β.
