Figure EV3. REL neuroblastoma cells are resistant to the cytotoxic effects of ionizing radiation despite incurring an equivalent increase in DNA damage.

Figure EV3

  1. DNA damage induced by 2 Gy ionizing radiation as measured by γ‐H2AX foci for SKNBE1 (DX, TP53 wild type) and SKNBE2C (REL, TP53 mutant). Fluorescence detection of γ‐H2AX foci and DAPI nuclear counterstain assessed, with representative images shown; SKNBE1 scale bar = 5 µm; SKNBE2C scale bar = 10 µm.
  2. Foci frequency plotted with mean shown for each condition in A (n = 42–58 nuclei/condition).
  3. Cell lysates from cells treated with 2 Gy of ionizing irradiation were immunoblotted for p53 and p53 target genes with β‐tubulin as a protein loading control.
  4. In vitro viability following ionizing radiation at 7 days; dotted line represents 50% viability. Data points are mean and SD error bars from triplicate wells, experiments are representative of at least three biological replicates.

Data information: Statistical analyses in B were performed using the two‐tailed Mann–Whitney U test, with significance P < 0.05.
