Effect of NLRP3-siRNA on thalamic pain. Baseline behavior tests were performed one day prior to siRNA/scramble injection into the VPL and VPM nuclei. Three days after the administration of siRNA or scramble, Coll IV or saline was microinjected into the VPL and VPM nuclei. Administration of siRNA significantly attenuated the increase in paw withdrawal frequency in response to (A) 0.07 g and (B) 0.4 g von Frey filaments and the decrease in paw withdrawal latency to (C) thermal and (D) cold stimuli at day 1, 3 and 5 following Coll IV microinjection on the contralateral side.
