Figure 3

Effects of unmodified and modified albumin on NW peptide binding to monocytes. (A) Unmodified albumin does not compete with the NW peptide binding. Monocytes were pre-incubated with various concentrations of unmodified human serum albumin (HSA) prior to the addition of NW peptide, staining, and analysis by flow cytometry. (B) Analysis of maleic-anhydride-modified albumin by SDS-PAGE. HSA and bovine serum albumin (BSA) were modified by maleic anhydride as described in the Section 4 and then analyzed by 10% SDS-PAGE followed by Coomassie blue staining. Unmodified HSA and BSA were included as controls. (C) Maleic-anhydride-modified albumin competes with the NW peptide binding to monocytes. Monocytes were pre-incubated with various concentrations of modified albumin before being stained with the NW peptide and analyzed by flow cytometry. The data shown in (A,C) are representative of at least eight independent experiments. m, Modified.
