Combined treatment with SR3029 and MG149 exhibits more potent effect on β-catenin acetylation, cell viability and proliferation in colon cancer cells. (A, B) SW480 (A) and HCT116 (B) cells were serum-starved for 12 h and subsequently treated with MG149 (5 and 10 μM) alone or combined with 240 nM SR3029 for 12 h The protein levels of β-catenin, acetylated β-catenin at K49, Tip60, CK1δ, CK1ϵ and GAPDH were detected by immunoblotting. (C–F) SW480 (C, E) and HCT116 (D, F) cells were serum-starved for 12 h and subsequently treated with MG149 (5 and 10 μM) alone or combined with 240 nM SR3029. After 48 h of treatment, MTT assay was used to detect cell viability (C, D). Cell proliferation was detected using BrdU incorporation assay (E, F).
