Effect of varying concentrations of test compounds on the growth of Toxoplasma gondii Type II (ME49) in human foreskin fibroblasts (HFF) cells. Equal amounts of freshly extracted tachyzoites of T. gondii (constitutively expressing luciferase protein - Fluc) were inoculated into HFF cells in culture and varying concentrations of compounds (A) JS-2-41 or (B) JS-2-44, were added immediately post-infection (green line). Control infected cells were treated immediately post-infection with volumes of DMSO equivalent to those used in the compound-treated cultures. Varying concentrations of ATQ added at the time of infection (red line) served as the positive control. The cultures were analyzed for parasite infectivity and proliferation after 48 h using a luciferase reporter gene assay. The luminescence generated by T. gondii luciferase activity was recorded and used to calculate the mean percent parasite inhibition values of each compound concentration relative to the DMSO-treated controls. Indicated values represent means from three independent experiments.
