Contribution of TbD1 on BCG Denmark to Copper sensitivity. (A) BCG Denmark WT (circles), ΔTbD1 (squares), and ΔTbD1:TbD1 (triangles) strains were grown at 37°C in standing Sauton’s liquid media, and CuSO₄ was added. Cultures were challenged with 0 μM CuSO₄ (black symbols) or 150 μM CuSO₄ (red symbols). Absorbance at 600 nm (OD₆₀₀) was measured for 2 weeks. The data are representative of three independent experiments. (B) Drop assay determining the Copper resistance of TbD1 mutants. Serial dilutions (10-fold) of BCG Denmark WT, ΔTbD1, and ΔTbD1:TbD1 cultures were spotted onto 7H11 plates containing 25, 100, or 150 μM CuSO₄. Data are representative of two independent experiments with replicates.
