The GC-box GGGCGG was bidirectionally scanned across the promoter region (−500,100) of 16,455 human genes in the EPD database using Motif Enrichment with a window size of 6 nucleotides and 1 nucleotide window shift. The relative abundance of the GC-boxes was plotted across the whole region. A sudden drop of the GC-boxes about 40 nucleotides upstream of TSS indicates the boundary between the proximal and the core promoter. A 100 nucleotide (−140, −41, red line) window was selected to ensure a high density of potentially functional GC-boxes.
