Mutant NDM-1 R45E R52E has lower ability to confer resistance and reduced in vivo protein stability under Zn(II) deprivation. (a) Cefotaxime MICs for E. coli DH5α cells expressing NDM-1 or NDM-1 R45E R52E, obtained in growth media supplemented with the indicated amounts of DPA and presented as relative to the corresponding MIC value obtained in 0 μM DPA. Data are presented as mean ± st. err. of 3 biological replicates. MIC values are presented in Table S2 in the Supplementary Information. (b) MBL protein levels in E. coli DH5α cells expressing NDM-1 or NDM-1 R45E R52E, quantified by immunoblot, at different time points after the addition of 200 μM DPA to the growth medium. Data points are presented as relative to control samples not treated with DPA, and correspond to the mean ± st. err. of 3 biological replicates. Data for each protein were fitted to a first-order exponential decay, indicated as a continuous curve. A representative immunoblot of these samples is presented in Figure S4 in the Supplementary Information.
