Effects of olaparib and PDD00017273 on cell cytotoxicity and PARylation activity in MC3T3-E1 cells. (A,B) Cell viability in olaparib- and PDD00017273-treated MC3T3-E1 cells. MC3T3-E1 cells were cultured in the condition of serum starvation for 24 h, and then treated with olaparib or PDD00017273 at concentrations of 0–30 µM in differentiation medium for 3 days. Cell viability was measured by CCK assay. (C) Intracellular PAR levels in MC3T3-E1 cells after treatment with olaparib and PDD00017273. MC3T3-E1 cells were treated with DMSO (solvent), 1 µM olaparib, or 1 µM PDD00017273 for 24 h, and then incubated in the absence or presence of 1 mM H2O2 for 10 min. Whole-cell extracts were analyzed by Western blotting using an anti-PAR antibody.
