Expression of L. donovani topoisomerase induces sensitivity to CPT-resistant yeast strain MBY3 (rad52Δ). Exponentially growing cells in dextrose, transformed with vectors carrying the leishmanial wild-type and different point mutants, driven by GAL1/GAL10 promoters, were serially 10-fold diluted starting from OD₅₉₅ = 0.3. Five microliters were spotted on selective media under repressed (2% dextrose) or induced (2% galactose) conditions, in the presence or absence of 15 and 30 μM CPT.
