The gel shows the specific pattern of cleaved bands of LdTopIB when exposed to 100 μM CPT with increasing amounts of protein, in contrast to its mutated counterparts, which exposition to CPT reveal how the drug is unable to stabilize cleavage complex in the presence of mutants LdTopIL^D353A/S and LdTopIL/S^N221S, showing no cleaved bands in the electrophoresis gel except of a unique band for LdTopIL/S^N221S.
