Docetaxel significantly enhanced Ad-mediated transgene expression independent of the levels of Ad binding receptors and virus replication. (A) CWR22rv and C4-2 cells were treated with vehicle or 10 nM docetaxel followed by infection with a panel of adenoviral vectors expressing EGFP, including replication-deficient Ads Ad5EGFP (fiber binding to CAR), Ad5/35EGFP (a hybrid Ad5/35 fiber binding to CD46) and Ad5RGDEGFP (a fiber-mutated by RGD), and replication-competent PRRA. The cells were harvested for flow cytometry 24 h after virus infection and the percentage of green fluorescent cells was determined. *p < 0.01. (B) The impact of docetaxel on the expression levels of CAR and integrins. CWR22rv and C4-2 cells were treated with vehicle or 2.5, 5 and 10 nM docetaxel for 24 h. The cells were lysed and the expression levels of CAR and integrin αν were determined by western blotting. The expression levels of integrin β1 and β3 were determined by flow cytometry. *p < 0.01, Δp > 0.05.
