Docetaxel significantly enhanced Ad-mediated transgene expression independent of the levels of Ad binding receptors and virus replication. (A) CWR22rv and C4-2 cells were treated with vehicle or 10 nM docetaxel followed by infection with a panel of adenoviral vectors expressing EGFP, including replication-deficient Ads Ad5EGFP (fiber binding to CAR), Ad5/35EGFP (a hybrid Ad5/35 fiber binding to CD46) and Ad5RGDEGFP (a fiber-mutated by RGD), and replication-competent PRRA. The cells were harvested for flow cytometry 24 h after virus infection and the percentage of green fluorescent cells was determined. *p < 0.01. (B) The impact of docetaxel on the expression levels of CAR and integrins. CWR22rv and C4-2 cells were treated with vehicle or 2.5, 5 and 10 nM docetaxel for 24 h. The cells were lysed and the expression levels of CAR and integrin αν were determined by western blotting.
