Disruption of mitochondrial thioredoxin system shortens lifespan of long-lived mitochondrial mutants. Deletion of trx-2 (A,B) or trxr-2 (C,D) significantly decreased the lifespan of nuo-6 and isp-1 worms but had little or no effect on the lifespan of wild-type worms. Disruption of trx-1 markedly decreased lifespan in nuo-6, isp-1 and wild-type worms (E,F). Deletion of trxr-1 significantly decreased nuo-6 lifespan but did not decrease the lifespan of wild-type worms (G). isp-1 trxr-1 double mutants could not be generated due to close proximity on the same chromosome. Statistical significance was assessed using a log-rank test. p-values indicate significance of difference between red and purple lines. A minimum of three biological replicates were performed.
