The effect of sortilin on apo(a) secretion is dependent on its carboxyl-terminal sorting motifs. HepG2 cells stably transfected with a 17-kringle form of apo(a) were transiently transfected with expression vectors encoding sortilin variants or the corresponding empty vector (pcDNA). The cells were subjected to a pulse-chase protocol followed by immunoprecipitation of medium and lysate samples using an anti-apo(a) antibody. A schematic representation of sortilin and the mutant variants used in shown in (A). Representative fluorographs are shown in (B). Densitometry was performed for lysate (C) and media (D) fluorographs. The data shown are the means ± SEM of at least three independent experiments.
