Dose- and incubation-time-dependent effect of RD2 on fractionated APP/PS1 mouse brain homogenate

DGC fraction 10 of APP/PS1 mouse brain homogenate, representing the peak of sFIDA signal, was diluted 1:2 and was incubated with 0, 5, 10, 20, or 50 μM RD2. Before analysis, samples were further diluted 1:5, resulting in a total dilution of 1:10, and the Aβ oligomer concentration was immediately analyzed by sFIDA assay. The final dilution factor during image acquisition was 1:20.

(A) Sections of representative TIRF images after 20 h of incubation. Red and green fluorescence channels were merged. The scale bar represents 10 μm.

(B) Aβ oligomer concentrations present in the original 1:2 diluted sample. Concentrations given here reflect the actual concentrations in the prepared sample after dilution and are not directly related to the concentrations in undiluted samples, shown in Figure 1.

Data are presented as mean ± SD ∗between groups, p < 0.05 (Kruskal-Wallis one-way ANOVA on ranks with Student-Newman-Keuls post hoc analysis). N = 3 (technical replicates).
