UBE-1099 induced nuclear factor erythroid 2–related factor 2 (Nrf2) activation through the inhibition of Kelch-like ECH-associated protein 1 (Keap1)-Nrf2 protein-protein interaction. (A) Chemical structure of UBE-1099. (B) Keap1-Nrf2 protein-protein interaction was measured by fluorescence polarization assay. (C) Plasma samples were collected at the indicated time points, and concentrations of CDDO-Im and UBE-1099 were measured by liquid chromatography tandem mass spectrometry. (D) Total RNA was isolated from renal tissues at the indicated time points. The level of the Nqo1 mRNA was measured and normalized to the level of Gapdh mRNA (internal control).
