Intracellular calcium amount under cisplatin stress of nimodipine pre-treated UB/OC−1 (a) and UB/OC−2 (b) cells. The 1 × 104 cells were seeded in black flat 96-well plates and treated as described in method section. Detection of intracellular calcium by the fluorescent dye Cal-520 showed a decrease by nimodipine pre-treatment in UB/OC−1 cells that was reduced with increasing cisplatin concentration. Examination of nimodipine-treated UB/OC−2 cells initially showed a decrease in the amount of calcium without stress induction (0.9% NaCl), which was minimally visible at 20 µM cisplatin and changed to an increase at 50 µM. The graphs show the results of three independent biological replicates. If the p values were <0.05 (* p < 0.05) compared to the solvent control cells (EtOH) were accepted as significant.
