Effect of storage of RBC under protein expression in monocytes. Monocytes (MØ) were co-cultured with RBC for 24 hours in four different conditions: RBC 0: MØ + 100% fresh RBC; RBC 21: MØ + 100% stored RBC for 21 days at four °C; Mix I: MØ + 80% fresh RBC + 20% stored RBC for 21 days at 4°C, and Mix II: MØ + 90% fresh RBC + 10% stored RBC for 21 days at 4°C Protein extracts were subjected to Western Blotting for quantification of (A) HO-1, (B) ferritin, and (C) Spic expression. The results are representative of four to seven different experiments. (D) Monocytes (MØ) were incubated with heme at concentrations of 3, 10, 30, and 60 μm. The protein extracts were submitted to Western Blotting for quantification of Spic levels. The results are representative of three different experiments.
