The MHC-II K225R DC defect is present in mice lacking T cells. Spleen DCs were isolated from RAG 1-deficient mice or MHC-II K225R mice crossed onto an RAGl-deficient background. (A) RAGl-deficient DCs (squares) or MHC-II K225R/RAGl-deficient DCs (circles) were cocultured with OT-II CD4+ T cells in the presence of increasing amounts of OVA protein. After 24 h, the amount of IL-2 secreted was determined by ELISA. The data shown are the mean ± SD obtained from three independent experiments. *p < 0.05.
