Schematic illustration describing the underlying mechanism of Toll‐like receptor 4 (TLR4)‐mediated microglial pyroptosis after spinal cord injury (SCI). After injury, overexpressed TLR4 recognises danger‐associated molecular patterns (DAMPs) and interacts with biglycan (BGN) to activate the JAK2/STAT1 pathway. Furthermore, STAT1, as a transcriptional activator, upregulates the expression of dead‐box helicase 3 X‐linked (DDX3X) and subsequently aggravates NLR family pyrin domain containing 3 (NLRP3) inflammasome activation and microglial pyroptosis.
