Amlodipine inhibits A549 cell proliferation by blocking the cell cycle G0/G1 phase. (A) Incubation of A549 with various concentrations (1, 2, 5, 10, 15, and 20 μM) of amlodipine (Aml) for 48 h. The effects of amlodipine on A549 cell proliferation were determined by MTT assay. (B) A549 cells were harvested 48 h after treatment with indicated concentrations of amlodipine, and apoptotic cells were measured by flow cytometry analysis. (C) Cells were pre-incubated with different concentrations of amlodipine for 48 h, and the induction of cell cycle arrest was detected by flow cytometry analysis. (D) The percentage of total cells at G0/G1, S, and G2/M phases. (E) The expressions levels of cell cycle-related proteins were determined by Western blotting after treatment with amlodipine.
