KPT-8602 inhibits activation of the NLRP3 inflammasome in vivo. (A) Substantia nigra compacta from PD mice administered KPT-8602 or vehicle were stained with an anti-Iba-1 and anti-ASC antibody to evaluate the activation of microglia and the NLRP3 inflammasome (5 mice per group). The number of Iba-1-positive cells (B) and ASC specks (C) was analyzed by ImageJ. Scale bars, 100 μm for low-magnification images and 50 μm for high-magnification images, respectively. (D) Expression levels of IL-1β, IL-6, and TNF-α in the SNc were detected by real-time PCR. The expression of Iba-1, pro-caspase-1, pro-IL-1β, cleaved caspase-1, cleaved-IL-1β, ASC, and β-actin in the SNc was analyzed by Western blotting (E), and the gray value of their bands was analyzed by ImageJ (F).
