Mechanism of SmiA-dependent LonA proteolysis of SwrA. In the absence of SmiA binding, SwrA remains in a protease-resistant state (unlicensed). Either constitutive or concurrent binding of SmiA and LonA to SwrA alters SwrA conformation to reveal a degron (licensed) that is then presented to the LonA in a protease-sensitive state for degradation. SmiA is then released and catalyzes multiple rounds of SwrA proteolysis.
