Antisense PPNAs targeting dnaB, ftsZ, and rpsH confer antibacterial activity against UPEC 536 at 1× MIC. (A) MICs for each 10mer PPNA, listed in Table 1, were plotted against the abundance (transcripts per million, TPM) of the respective PNA’s target. Grey shaded area highlights the four PPNAs with the strongest antibacterial efficacy against UPEC 536. (B, C) Bactericidal effects were determined for (KFF)3K-dnaB (5 μM), (KFF)3K-ftsZ (2.5 μM) and (KFF)3K-rpsH (2.5 μM) at 1× MIC against UPEC 536 (105 cfu/ml). Exposure to an equal volume of water served as control (black). Samples were taken at the indicated time points post treatment. (B) Spot assays with serial dilutions (10–1 to 10–3) and (C) cfu evaluation on LB plates was performed to determine the number of viable cells. Curves represent the mean of the biological replicates. Each experiment was performed twice.
