Construction of pTnHB100. The minitransposon TnHB100 was obtained by cloning the hup region from cosmid pAL618 into a pUT derivative (pCAM140) as described in Materials and Methods. It comprises a 21,376-bp DNA region located between the 19-bp Tn5 inverted repeats, represented by solid boxes. The thick line downstream from the hup hyp region indicates DNA from the vector pLAFR1. The shaded bar over the gene cluster represents the restriction fragment used for deletion of the spectinomycin resistance gene (see Fig. 4).
