Southern blot analysis of DNA isolated from Thermoanaerobacterium strain YS485 wild type and YS485 transformed with suicide vector pUXK. The DIG-labeled probe against the 5′ end of xynA was generated as described in Materials and Methods. Plasmid pUXK contains neither NsiI nor NcoI but one PstI recognition site. The probe hybridized with the PstI-linearized pUXK.
