The blocking activity of fisetin on apoptosis and survival inhibitory effects was counteracted in H2O2-treated ARPE-19 cells. Cells treated with 20 μM fisetin and/or 5 μM ZnPP for 1 h were further treated with 500 μM H2O2 for 24 h (A,B) Flow cytometry with annexin V/PI dual staining was performed. Representative histograms (A) and quantitative analysis (B) are shown. (C) Cell viability was measured by the CCK8 analysis for cells cultured under the same conditions. The blocking effect of fisetin on H2O2-induced apoptosis was significantly counteracted by ZnPP, and the cytotoxic protective effect of fisetin was also significantly moderated.
