Doubly labeled apo hAgo2^N/PAZ (A) and binary hAgo2^N/PAZ-guide RNA complexes (B) were immobilized via an hAgo2-directed antibody, whereas ternary hAgo2^N/PAZ-guide RNA-target RNA complexes (C) were immobilized via the target RNA^long-Biotin. In case of measurements with miRNA substrates used in (D) (guide RNA^p-miR451a and target RNA^osr1) and (E) (pre-miR451a) the hAgo2^N/PAZ complexes were immobilized using an hAgo2-directed antibody. All FRET efficiency histograms show data of at least three independent measurements. All data were fitted using a Gaussian equation. Source data provided as a Source Data file.
