3H-2-PAM@scFv-Lipo was intravenously administered to C57BL6 mice at a dose corresponding to 5 mg/kg of liposomally-conjugated scFvs, and 3.4–4.3 × 107 DPM/kg of 3H-2-PAM (n=3). An aliquot of orbital vein blood sample was collected at 0.5, 2, 15, 30, 60, 120 and 240 min after injection. Plasma amounts of 3H-2-PAM were determined by scintillation counting and used to determine the AUC. After 4 hours of circulation, mice were whole body perfused, organs were isolated, and scintillation counting was used to measure 2-PAM uptake in the whole organ homogenates. Organ uptake parameters were calculated as described in the Materials and Methods. Reported are means ± S.D., n=3 independent mice.
