WT or Clk1+/− mutant mice were sacrificed and striatum, hippocampus and prefrontal cortex tissues, together with LV-shNC and LV-shClk1 PC12 cells, were used as follows: a, b ICP-MS analysis of total iron levels; c, d mRNA expression of DMT1, TfR1, FTL and Hepcidin; e Western Blotting for HIF-1α, FPN1, FTL and Clk1; f, g ELISA assay of hepcidin; h Western Blotting for HIF-1α, FPN1 and FTL. Statistical analyses for a, c, d, e, g and h were performed using two-way ANOVA followed by Bonferroni-corrected tests and statistical analyses for b and f were performed using Student’s t test (*P < 0.05, **P < 0.01, ***P < 0.001).
