The gating and sorting strategy that preceded the single cell sort that led to the identification of the CO3 TCR from healthy donor CO3 (a) or the CO22 TCR from healthy donor CO22 (b) is shown. The CO3 or CO22 TCR was transfected into 293 T cells, together with the CD3 complex, and stained with CD1a, CD1b, CD1c, or CD1d tetramers and an antibody against CD3 (c). Results in c are representative of 2 experiments. d The unmodified CO3 TCR was transfected, or its Vγ4 γ chain was replaced with the indicated Vγ chains from CD1b or CD1d-specific γδ TCRs, or its Vδ1 chain was replaced with the Vδ1 chain of the CD1b-specific BC14.1 TCR. The indicated tyrosine residue at position 104 of γTCR chain was present in CO3 and DP10.7. The transfected cells were stained with CD1a tetramer (top row) or an antibody against the γδ TCR.
