Nitrate utilization promotes S. Typhimurium systemic infection in the mice. (a) Drinking water containing 20 g/L of sodium nitrate was provided for mice from 3 days before i.p. infection to the end of the experiment. Meanwhile, the drinking water without the addition of sodium nitrate was used as control. Bacterial burdens of liver and spleen in infected mice were assessed at day 5 post-infection. n = 6 mice/group. (b) Survival curves for mice infected i.p. with the S. Typhimurium WT, ΔnarK, ΔhmpA, or ΔhmpAΔnarK, n = 10 mice/group. (c) Liver and spleen bacterial burdens in mice infected with the S. Typhimurium WT, ΔnarK, ΔhmpA, or ΔhmpAΔnarK at day 5 post-infection. n = 6 mice/group. (ac) Data were combined from two independent experiments and are presented as mean ± SD. p values were determined using Mann–Whitney U test (a,c) or log-rank Mantel–Cox test (b) (* p < 0.05; ** p < 0.05; **** p < 0.0001).
