Salt alters the degree of methylation in loosely bound pectins. (A) Seven-day-old Arabidopsis Col-0 seedlings were treated for 1, 3 and 6 h with water (mock, ‘m’) or 100 mM NaCl (‘N’). From the AIRs, 5 μg sugars were extracted and treated with double-distilled water (native) or 0.1 M Na2CO3, pH 11, to obtain de-methylated HG (deM-HG). Dot blots was performed using 2F4 (n=3). (B) Five micrograms of sugars extracted from 6-day-old seedlings after 24 h treatment with water (m) or 100 mM NaCl (‘N’) were used to perform dot blots as in A.
