Proposed pathogenesis of hypomagnesemia due to R480L of CNNM2. (A) Normally, Mg²⁺ is effluxed after Mg²⁺-ATP binds to wild-type CNNM2 under magnesium depletion. (B) Mutation in CNNM2 yields a decreased binding force between CNNM2 and Mg²⁺-ATP and subsequent hypomagnesemia under magnesium depletion.
