THP-1 macrophages were transduced with Adv-FoxO1-ADA and Adv-Empty adenoviruses in culture medium. After 24-hour incubation, THP-1 macrophages were analyzed by real-time qRT-PCR assay. (I) Macrophage mRNA levels in human primary macrophages. Human primary macrophages isolated from liver biopsies of deidentified human patients were transduced with Adv-FoxO1-ADA and Adv-Empty adenoviruses in culture medium. After 24-hour incubation, human macrophages were analyzed by real-time qRT-PCR assay. Data were obtained from at least 3 independent experiments, and are expressed as mean ± SEM. Statistical analysis in G was performed using 1-way ANOVA with Tukey’s multiple-comparison test, in H using a 1-tailed, unpaired t test, and in I using a 2-tailed, unpaired t test. *P < 0.05, **P < 0.01, ***P < 0.001.
