Measurement of ciliary beating in the adult telencephalon

(A) Diagrams presenting the overall procedure to measure ciliary beating in the brain explant using a light transmission microscope. (A1) Oxygenated artificial cerebrospinal fluid (aCSF) is delivered to the brain explant through a perfusion system (inlet is labeled as “IN” and outlet as “OUT”). (A2) A region of interest is identified using a 40× objective prior to the recording (A3). The pink areas indicate the regions where ciliary beating can be observed (the tela choroida and choroid plexus). The big red box on the right side indicates high magnification of the small red box in the left side. The clusters of magenta lines display motile cilia in the explant. (A4) The analysis can be performed using a MATLAB script or the FreQ plugin in ImageJ/Fiji.
