Schematic representation of the fluctuation analysis to determine an accurate LOH frequency. For a given strain in one growth condition, 24 parallel cultures are generated. For each parallel culture, 8 FACS analyses of 106 cells are conducted for a total sampling size of 8 × 106 cells. A total number of mono-fluorescent cells is calculated per parallel culture. The median LOH frequency (+/−SD) is assessed using the 24 parallel cultures.
