Overlapping transcription promotes HilD-dependent bistability of SPI-1 expression. Strains MA14358 (hilD+) and MA14569 (hilDΔ309) carry the tetR-PtetA cassette (Fig. 3A) combined with hilA::gfpSFΔK28 and the chromosomal PTac-mCherry in the ARA-inducible-NusG depletion background. Cells grown to early stationary phase under the indicated conditions were used for single-cell analysis by flow cytometry.
