Figure 8

Plastid RNA accumulation in the WT and nadk2-2. A, RNA gel blot analysis of the psaA-psaB-rps14 gene cluster in WT and nadk2-2 plants. Twenty micrograms of total RNA from WT and nadk2-2 mutant plants were size-fractionated by agarose gel electrophoresis, transferred onto a nylon membrane, and probed with labeled cDNA probes. The genetic composition of this gene cluster and the locations of probes used for hybridization are shown above. Distinct transcription initiation sites of the plastid- (PEP) and nucleus-encoded (NEP) polymerases are indicated by arrows. The rRNAs stained by ethidium bromide (EtBr) are shown on the right of the blots as loading control. B, RNA gel blot analysis of other plastid genes in WT and nadk2-2 plants. The experimental procedure was the same as in (A).
