Sirtuin 3 and ROS. (A); Sirt3 stimulates the expression of key antioxidant enzymes MnSOD2 and catalase via deacetylation of FOXO3a. Deacetylation occurs either in the mitochondria with subsequent translocation of FOXO3a into the nucleus or by means of nuclear fraction of Sirt3 in the nucleus. (B); Sirt3 directly deacetylates and activates mitochondrial enzymes implicated in ROS quenching (IDH2 and SOD2). (C); Sirt3 decreases electron leakage from mitochondrial electron transport chain by deacetylation of its complexes. (D); The antioxidant activity of Sirt3 leads to HIF1 destabilization and Ras-signaling suppression.
