Ectopic expression of miR-144/451 downregulates DCs function. Mice splenic DCs were infected with lentiviruses for 24 h, the frequency of DCs (A) and expression of CD80 (B), CD86 (C), CD40 (D) and TNF-α (E) on DCs was detected. CD8⁺T cells were co-cultured with lentiviruses infected DCs for 24 h, surface markers CD69 (F), NKG2D (G) and intracellular cytokines IFN-γ (H), TNF-α (I) were detected in CD8⁺T cells. Human PBMCs were cultured in presence of GM-CSF (50 ng/ml) and IL-4 (50 ng/ml) for 5 days. Expression of CD80 and TNF-α (J, K) by monocyte-derived DCs were detected after 48 h lentiviruses infection.
